Characterization of candidate genes in congenital heart disease

Congenital heart disease (CHD) is a complex disorder which is estimated to affect nearly 1 % of newborns. Using genome-based screening methods we have identified several genes which may be involved in CHD. In my phD-project I will use digital expression profiling of human embryonic hearts to identify novel candidate genes in CHD. CHD candidate genes will be screened for mutations in a cohort of patients and the genes will be characterized functionally. The spatial and temporal expression of candidate genes during human heart development will be analyzed using QPCR, immunohostochemistry and in situ hybridization. The P19CL6 cell model will be used to study the functional effect on cardiomyogenesis caused by RNAi mediated knock-down of candidate genes. Finally, functional effects on cardiac development caused by candidate gene knock-down will be analyzed in vivo using a zebrafish model
